I want to set my ports of my B400p on point to multipoint mode. Because i have mutiple ISDN.

you can set that to PTMP mode in misdn.conf:
<port mode="te" link="ptmp">1</port>.

We use Elastix, and its reloading the old defaults every time when i change it.

When i do asterisk -r

misdn show port 1
misdn show port 2
misdn show port 3
misdn show port 4
